玉屏风散合四物汤对慢性阻塞性肺疾病大鼠气道7种细菌的影响

             

摘要

目的 观察玉屏风散合四物汤对慢性阻塞性肺病(COPD)大鼠气道肺炎克雷伯菌、铜绿假单胞菌、流感嗜血杆菌、肠杆菌、链球菌、奈瑟菌、乳酸杆菌的影响作用,为玉屏风散合四物汤治疗COPD提供一定的实验依据.方法 按随机数字表将50只SPF级雄性Wistar大鼠分为正常组,模型组,玉屏风散合四物汤高、中、低剂量组,每组各10只.通过气管滴注脂多糖加烟熏的方法建立COPD大鼠模型.造模结束后,给药组分别给予不同浓度玉屏风散合四物汤灌胃,正常组和模型组以无菌蒸馏水灌胃,每日1次,连续30 d.收集所有大鼠肺组织匀浆进行各种细菌的高通量测序检测,取左肺组织制作病理切片.结果 ①与正常组相比,模型组乳酸杆菌含量显著降低,其余6种细菌含量均显著升高,差异有统计学意义(P<0.05);②与模型组相比,玉屏风散合四物汤高、中、低剂量组的肺炎克雷伯菌、铜绿假单胞菌含量显著降低,高剂量组的流感嗜血杆菌、肠杆菌、链球菌、奈瑟菌含量均显著降低,差异有统计学意义(P<0.05),而中、低剂量组差异无统计学意义(P>0.05);③与模型组相比,玉屏风散合四物汤低、中剂量组的乳酸杆菌含量显著升高,差异有统计学意义(P< 0.05),而高剂量组差异无统计学意义(P>0.05).结论 玉屏风散合四物汤可使COPD大鼠肺炎克雷伯菌、铜绿假单胞菌、流感嗜血杆菌、肠杆菌、链球菌、奈瑟菌含量明显减少,乳酸杆菌含量增多,这可能是其治疗COPD的重要机制之一.%Objective To observe the effect of Yupingfeng San and Siwu Decoction on Klebsiella pneumoniae,Pseudomonas aeruginosa,Haemophilus influenzae,Enterobacter,Streptococcus,Neisseria and Lactobacillus in rats with chronic obstructive pulmonary disease (COPD),in order to provide experimental basis for Yupingfeng San and Siwu Decoction in the treatment of COPD.Methods Fifty SPF male wistar rats were divided into 5 groups,incliding normal group,model group,Yupingfeng San and Siwu Decoction high dose,middle dose and low dose groups,with 10 cases in each group.The COPD rat model was established by intratracheal instillation of lipopolysaccharide plus smoked methods.After molding,the rats in the dose groups were given to different concentrations of Yupingfeng San and Siwu Decoction by gavage,while the rats in the normal group and model group were fed with sterile distilled water,once time a day for 30 days.All the lung tissues of rats were collected,to for high-throughput sequencing of various bacteria,and the left lung tissue was used to make pathological sections.Results ①Compared with the normal group,the content of Lactobacillus in the model group was significantly decreased,and the contents of the other six bacteria were significantly increased,with statistically significant difference (P < 0.05).②Compared with model group,the contents of Klebsiella pneumoniae,Pseudomonas aeruginosa in the Yupingfeng San and Siwu Decoction high,middle and low dose groups were significantly reduced,the contents of Haemophilus influenzae,Enterobacter,Streptococcus,Neisseria in the high dose group were significantly reduced,with statistically significant difference (P < 0.05),but there was no significant difference between the middle and low dose groups (P > 0.05).③The contents of Lactobacillus in the Yupingfeng San and Siwu Decoction low and middle dose groups was significantly increased,compared with model group,with statistically significant difference (P < 0.05),there was no significant difference between high dose group and model group (P < 0.05).Conclusion The coments of Klebsiella pneumoniae,Pseudomonas aeruginosa,Haemophilus influenzae,Enterobacter,Streptococcus and Neisseria can be significantly reduced in rats with COPD by Yupingfeng San and Siwu Decoction,the content of Lactobacillus increasing,which may be one of the important mechanisms for the treatment of COPD.
