多形性日光疹患者外周血单个核细胞趋化因子受体CXCR1的检测

Objective: To determine the level of chemokine receptors CXCR1 in patients with polymorphous light eruption.Methods: Real - time quantitative PCR technique was applied to quantitatively analyze the expression of CXCR1 mRNA in peripheral blood mononuclear cells (PBMC), the CXCR1 expression in PBMC was detected by flow cytometry and serum level of IL- 8 measured by double antibody sandwich enzyme - linked immunosorbent assay (ELISA) in 35 cases of polymorphous hght eruption and 30 healthy controls.Results: The CXCR1 mRNA level in PBMC from cases of polymorphous light eruption was 7.59± 1.21, significantly higher than that in normal controls (3.63±0.98) (P ＜0.001).The CXCR1 expression in PBMC of the patients was 42.28 ± 12.79, significantly higher than that in normal controls (22.63 ± 10.94 ) ( P ＜ 0.001 ).The serum level of IL - 8 in the patients was 2.74 ± 0.58, which were also markedly higher than that in normal controls ( 1.63 ± 0.58 ) ( P ＜ 0.01 ).Conclusion: The increased IL- 8 and its receptor CXCR1 may be involved in the pathogenesis of polymorphous light eruption.%目的:检测CXC型趋化因子受体CXCR1在多形性日光疹患者外周血单个核细胞中的表达.方法:应用实时定量PCR法检测35例多形性日光疹患者和30名健康对照者外周血单个核细胞CXCR1 mRNA的表达,用流式细胞仪分析外周血白细胞CXCR1的表达,用ELISA法检测血清中IL-8的水平.结果:多形性日光疹患者外周血单个核细胞中CXCR1 mRNA表达水平为7.59±1.21,明显高于对照组3.63±0.98(P<0.001),CXCR1的表达率为(42.28±12.79)%,明显高于对照组(22.63±10.94)%(P<0.001);血清中IL-8的水平为2.74±0.58,显著高于对照组1.63±0.58(P<0.01).结论:CXC型趋化因子IL-8及其受体CXCR1在多形性日光疹患者外周血中的水平显著增高,可能参与了多形性日光疹的发病.
