柞蚕丝素蛋白/羟基磷灰石支架的制备及生物学特性检测

         

摘要

目的:制备柞蚕丝素蛋白/羟基磷灰石复合支架,改善单纯羟基磷灰石(HA)支架材料的生物学特性。方法:将羟基磷灰石多孔支架材料浸泡在柞蚕丝素蛋白的水溶液中,抽真空处理48h,采取冷冻干燥法制备的方法,进行羟基磷灰石支架表面修饰。实验分为HA 支架组(单纯羟基磷灰石支架组)与(TSF/HA)柞蚕丝素蛋白/羟基磷灰石支架组。按ISO 10993标准,两组分别做体外降解实验、吸水测试及细胞粘附实验。结果:SEM镜下观察TSF/HA支架孔隙内壁上细胞粘附高于单纯HA支架,吸水率实验检测TSF-HA支架组吸水率高于单纯HA支架组,体外降解实验TSF/HA支架降解速率快于单纯HA支架,各项检测差异均有统计学意义(P<0.05)。结论: TSF/HA支架材料具有比单纯HA支架材料更优良的生物学特性,可以用于后续的实验研究。%Objective:Prepare the artificial scaffold with tussan silk fibroin(TSF) and hydroxyapatite(HA), improve the biological properties of simple hydroxyapatite. Methods:The porous scaffolds were soaked in TSF water solution 24hours, then these scaffolds were vacuum treatment at-10℃for 48 hours and the complex scaffold----TSF/HA were gained. The scaffolds were separated into two groups, simple HA and complex TSF/HA. According to the ISO 10993 criterion,the biological properties detection included degradation in vitro, water absorption test and celladhesion test. Result:The complex TSF/HA scaffold had innumerable TSF bal on the inner wal of cells of HA under SEM. The degradation and water absorption of complex TSF/HA scaffold were obvious fast than that of simple HA (P<0.05). The celladhesion of complex TSF/HA were significantly higher than simple HA scaffold (P<0.05). conclusion:Complex TSF/HA scaffold had bet er biological properties than HA, which could be used in the subsequent experimental studies.
