三种猪病相关microRNAs的研究

     

摘要

本实验旨在探索繁殖与呼吸综合征(PRRS)、口蹄疫(FMD)和传染病胃肠炎(TGE)感染猪的microRNA (miRNA)变化,为疫病监测提供新途径。采用生物信息学方法预测可能的相关病毒miRNA,在MARC-145细胞系中转染10种miRNA mimic和inhibitor。相关实验结果发现过表达miR-125b、miR-147和miR-181降低了PRRSV的繁殖,而敲低miR-125b、miR-147和miR-181能促进PRRSV复制;感染猪口蹄病毒的样本中let-7a表达量明显上升,而miR-146b表达量明显下降;对照组中miR-142的表达量高于患有传染性胃肠炎样品。说明相关microRNAs可能参与病毒调控,可用作检测参考指标。%The study was aimed to explore the pig microRNA(miRNA)associated with porcine reproductive and respiratory syndrome virus(PRRSV),foot and mouth disease virus(FMDV)of pig and transmissible gastroenteritis virus (TGEV)infections in order to explore new means for disease detection. At first,the virusassociated miRNAs were predicted by bioinformatics method. Then,MARC-145 cells were transfected with the mimic or inhibitor of various miRNAs,and the related experiments showed that over expression of miR-125b,miR-147 and miR-181 could inhibit PRRSV replication and their knockdown could promote virus replication;the let-7a expression was increased signifi cantly while the miR-146b expression decreased signifi cantly in FMD-infected pig samples;and the miR-142 expression in the control group was higher than in the TGE-infected samples,suggesting that associatedmiRNAs were possibly involved in virus modulation and could be used as disease indicators.
