紫外诱变复合重金属离子抗性突变选育螺旋霉素高产菌株

         

摘要

以螺旋霉素链霉菌(Streptomyces spiramyceticus)YL0113-01为出发菌株,采用紫外线诱变,并以重金属Co2+为筛选因子,获得一株遗传稳定且螺旋霉素发酵效价比出发菌株提高6%的菌株。再对该菌株采用亚硝基胍(NTG)诱变,以缬氨酸结构类似物 L-α-氨基丁酸作为筛选因子得到一株发酵效价较出发菌株提高14%的菌株。%A parent strain Streptomyces spiramyceticus YL0 1 1 3-0 1 was treated by UV and using heavy met-al Co2+ as a screening factor.A stable strain was obtained with fermentation titer increased by 6% than parent strain.Then the mutant was treated by NTG with L-α-amino-butyric acid(an analogue of valine)as a screening factor,and a strain with fermentation titer increased by 1 4% than parent strain was obtained.
