雄黄染毒后大鼠脑组织中氨基酸类神经递质含量的变化

     

摘要

研究了雄黄对大鼠脑组织氨基酸类神经递质含量的影响.将32只Wistar大鼠随机分为对照组(0.5%CMC-Na)以及低剂量(0.3 g/kg)、中剂量(0.9 g/kg)、高剂量(2.7 g/kg)雄黄染毒组,通过连续灌胃给予雄黄混悬液两周.采用高效液相色谱-柱前衍生化法测定了大鼠脑组织中氨基酸类神经递质含量的变化.结果表明,与对照组比较,低剂量组大鼠脑组织中丝氨酸、甘氨酸和γ-氨基丁酸含量明显增加.中、高剂量组大鼠脑组织中同型半胱氨酸、谷氨酰胺、丝氨酸和天冬氨酸含量明显比对照组的低.总体而言,雄黄可对大鼠脑组织氨基酸类神经递质产生影响,氨基酸类神经递质可能是雄黄毒性作用的靶点之一.%The effect of realgar on the content of amino acid neurotransmitters in brain tissues of rats was investigated. 32 Wistar rats were randomly categorized into control, low dosage, moderate dosage, and high dosage realgar treated groups. The realgar treated groups were treated with realgar by gastric perfusion at a dosage of 0. 3 g/kg, 0. 9 g/kg, and 2. 7 g/kg, and the control group were orally given the same volume of 0.5% sodium carboxymethylcellulose (CMC-Na) for 2 weeks. The content of amino acid neurotransmitters in the brain tissues of the rats was determined by means of high performance liquid chromatography in association with precolumn derivatization. Result shows that the levels of serine, glycine and y-aminobutyric acid in low dose realgar treated group are significantly elevated as compared with the control group; while the levels of aspartate, homocysteine, glutamine and serine in moderate and high dose realgar treated groups are obviously decreased as compared with the control group. In one word, realgar has effect on amino acid neurotransmitters in brain tissues of rats, and amino acid neurotransmitter may be one of the toxic targets of realgar.
