纸浆蔗渣酶法水解反应过程的考察

         

摘要

以蒸煮时间5.5小时,温度为155~160℃,P=0.6MPa, pH为4.5的40目的纸浆蔗渣为底物,日本Yakult生物化学试剂公司的Onozuka-R-10纤维素酶为催化剂,在50℃,pH为4.8(乙酸—乙酸钠缓冲液)的条件下,考察了釜式和固定床反应器对酶解反应过程的影响,测定了在不同底物浓度时的酶解反应还原糖得率、探讨了酶的吸附以及酶活性在酶解反应过程的变化情况。实验结果表明:在本实验条件下,底物浓度高,转化率则低;固定床反应器对酶的吸附比釜式反应器略多;酶的活性变化在两种反应器型式中几乎没多大差别。%Paper pulp bagasse celluloses were pretreated under the conditions of τ=5.5h, T=155-160℃,P=0.6MPa,pH=4.5. The size of paper pulp bagasse celluloses was 40 meshes. The Yakult 0nozuka R-10 cellulase was used in the experiments. The paper pulp bagasse cellulose enzymatic hydrolysis were carried out in a tank reactor and/or a fixed bed reactor under the conditions of T=50℃, pH=4.8. The hydrolysis reducing sugar yields, enzyme adsorptivity and activity were inspected for different cellulose concentrations. The experiment results indicated that the higher bagasse concentration, the lower reducing sugar yields,and the enzyme adsorption amount were more in the fixed bed reactor than in the tank reactor in the experimental conditions. The enzyme activity changes with hydrolysis were closed in the two type reactors. The enzyme activity decrease was relatively quick in the first 20hr. It was experimentally observed that enzyme always loses its entire activity when the reaction periods exceed 140hr (close 6 days).
