超声波辐射强化污泥蛋白质的提取

     

摘要

为了提取污泥胞内蛋白质,以CaO为药剂,对污泥进行了热搅拌和超声波辐射的对比实验,考察了超声波发生器输出端电流强度、作用时间、pH值对蛋白提取率的影响,通过正交试验对实验条件进行了优化,并采用SDS-PAGE对提取出的蛋白质的相对分子质量进行了分析.结果表明:超声波辐射提取污泥蛋白质的最佳工艺条件是:辐射时间20 min,pH=12,电流强度0.7 A,在此条件下蛋白质提取率可达73%左右,相对分子质量介于32.942×10<'3>-569.55×10<'3>.因此,超声波辐射替代热搅拌应用于污泥蛋白质的提取是一种切实可行的方法.%In order to extract the sludge intracellular proteins, the contrast experiment of sludge by ultrasonic radiation and thermal stir was performed with calcium oxide as reagent.The effects of the output current intensity of ultrasonic generator, process time and pH value on the protein extraction yield were investigated.The experimental conditions were optimized by orthogonal test, and the relative molecular mass of the extracted proteins was analyzed by SDS-PAGE.The results show that the optimal process conditions for the extraction of sludge proteins by ultrasonic radiation are as follows: radiation time 20 min, pH = 12, and output current intensity 0.7 A.Under such conditions, the proteins extraction yield can reach about 73%, and relatively molecular mass is between 32.942 ×103-569.55 × 103.Consequenfiy, ultrasonic radiation in stead of thermal stir is practical for the extraction of sludge intracellular proteins.
