针刺对应激性胃黏膜损伤大鼠血浆和下丘脑中β-内啡肽含量的影响

         

摘要

Objective To explore the mechanism of acupuncture in preventing and treating stress-induced gastric muco-sal injury from the view of brain-gut axis. Methods Forty male Wistar rats were randomly divided into normal control, model 1, treatment, model 2 and prevention groups (n = 8). Gastric mucosa injury model was established by-intragastric perfusion of dehydrated alcohol (1.0 mL/rat). Rats of the treatment group were treated with acupuncture after modeling, while those of the prevention group treated first, followed by modeling. The time of modeling in model 1 group and model 2 group was simultaneous with that of the treatment group and prevention group respectively. Acupuncture was applied to "Zusanli"(ST 36) ,"Zhongwanw(CV 12) and "Neiguan"(PC 6) for 20 min , once daily for 5 days. Before sampling the tissues, 10% charcoal suspension was intragas-tric perfused 1 h for analyzing the rate of gastrointestinal propulsionC distance from the upper end of the charcoal powder to the car-dia / total length of the cardia to the anus ×100%). Gastric mucosal ulcer index was measured by using Guth's method. The contents of β-endorphin(β-EP)in plasma and hypothalamus were determined by enzyme-linked immunosorbent assay. Results Compared with the normal control group, the gastrointestinal propulsion rates were decreased considerably in the two model groups (P<0. 05), while the gastric mucosal ulcer indexes and the contents of p-EP in both plasma and hypothalamus were increased significantly in the model groups (P<0. 05, P<0. 01). Compared with the corresponding model groups, the gastrointestinal propulsion rate was increased remarkably in the prevention group (P<0. 05), and the gastric mucosal ulcer indexes and thecontents of plasma β-EP level were decreased obviously in both treatment and prevention groups (P<0. 05, P<0. 01). The contents of hypothalamic β-EP were increased further in the later two groups (P<0. 01). Conclusion Acupuncture of ST 36, CV 12 and PC 6 can promote the repair of gastric mucosal injury and improve gastrointestinal function, which may be related to its effects in reducing plasma β-EP and upregulating hypothalamic β-EP level. Acupuncture also has a better effect in preventing gastric mucosal injury.%目的:从脑-肠轴角度探讨针刺在预防和治疗应激性胃黏膜损伤方面的作用机制.方法:将雄性Wistar大鼠随机分为正常组、模型1组、模型2组、治疗组、预防组,每组8只.采用乙醇灌胃法造成大鼠应激性胃黏膜损伤模型.治疗组先造模后针刺,预防组先针刺后造模;模型1组和模型2组的造模时间分别同治疗组和预防组.穴选“足三里“中脘”“内关”,针刺干预5d.实验结束前1h各组用10%炭粉混悬液灌胃,然后取材,计算炭粉推进百分率(胃肠推进百分率),肉眼观察大鼠的胃黏膜形态,Guth法计算胃黏膜损伤指数,酶联免疫法测定血浆和下丘脑中β-内啡肽的含量.结果:与正常组比较,两个模型组的胃肠推进百分率均明显降低(P<0.05),肉眼可见胃黏膜损伤明显,损伤指数明显升高(P<0.05,P<0.01),血浆及下丘脑中β-内啡肽含量均明显升高(P<0.01);与相应的模型组相比,预防组的胃肠推进百分率显著提高(P<0.05),治疗组和预防组大鼠的胃黏膜损伤程度明显减轻,损伤指数明显降低(P<0.05,P<0.01),血浆中β-内啡肽含量明显降低(P<0.05,P<0.01),下丘脑中β-内啡肽含量显著升高(P<0.01).结论:针剌能促进胃黏膜损伤的修复,提高胃肠功能,与其降低血浆中β-内啡肤含量,增加下丘脑中β-内啡肽含量有关.针刺“足三里”中脘”“内关”预防胃黏膜损伤的效果明显.
