杯状细胞及5-HT、SP-IR细胞在药物依赖性便秘大鼠盲肠中的表达及意义

         

摘要

目的 观察药物依赖性便秘大鼠盲肠杯状细胞、5-羟色胺免疫反应(5-HT-IR)细胞、P物质免疫反应 (SP-IR) 细胞的形态分布及数量的变化,探讨大鼠盲肠杯状细胞及5-HT-IR、SP-IR细胞在药物依赖性便秘的可能作用.方法 将45只SD大鼠均分为正常对照组、大黄模型组和酚酞模型组,两模型组分别用大黄和酚酞灌胃建立大鼠药物依赖性便秘模型,用普通组织学、免疫组织化学SABC法和细胞计数进行研究大鼠盲肠杯状细胞、5-HT-IR细胞及SP-IR细胞所发生的形态分布及数量的变化.结果 与正常对照组比较,两模型组盲肠杯状细胞、5-HT-IR细胞数量明显减少(P<0.05),且免疫染色减弱.结论 盲肠杯状细胞及5-HT-IR细胞的改变也可能是药物依赖性便秘的一个重要因素.%Objective To study possible functions of the drug-dependent constipation by observing morphological distributions and quantitative changes of goblet cells and 5-HT and SP immunoreactive cells of appendix of the drug-dependent constipation in rats. Methods 45 SD rats were randomly divided into the normal control group, rhubarb model group and phenolphthalein model group 15 SD rats in each group. To develop model of drug-dependent constipation in rats by infusing stomach with rhubarb and phenolphthalein in two model group rats and to use the immu-nohistochemistry SABC, common histological method and cell counting for observing the morphological distributions and quantitative changes of goblet cells, 5-HT and SP immunoreactive cell of appendix in rats. Results Compared with the normal control group, the number goblet cells and 5-HT of intestine of rats decreased obviously( P < 0. 05 ) and the immunohistochemical reaction duoenum deceased in latter. Conclusion It is suggested that the changes of morphological distribution and quantity of appendix goblet cells, immune cells 5-HT may be an important factor of drug dependence constipation.
