Objective To explore the proteins expression and significance of β-catenin, DKK-1 in bisphenol A ( BPA) exposure before and after birth on male rat reproductive cells. Methods ICR infected pregnant rats drink-ing water from the conception 0 day(PGD0)were randomly divided into:BPA solvent control group, 10 nmol/L,50 nmol/L,300 nmol/L BPA groups. Male young rats were killed in the 6th week(PGD42)after birth,β-catenin and DKK-1 protein expressions in testicular tissue were detected by immune-histochemical stained and Western blot. Re-sults Compared with the solvent control group, testicular viscera coefficient in BPA exposure group young mice decreased. Compared with the solvent control group, β-catenin and DKK-1 protein expression in BPA exposure group significantly increased with immunohistochemical results and Western blot. Conclusion The main proteins ofβ-catenin, DKK-1 in Wnt/β-catenin signaling pathway proteins may be involved in the BPA exposure before and after birth on male rat reproductive cells.%目的:探讨 Wnt/β-catenin 信号通路主要蛋白(β-catenin、DKK-1)在出生前后双酚A(BPA)暴露的成年子鼠雄性生殖细胞中的表达及意义。方法自受孕第0天(PGD0),ICR孕鼠饮水BPA染毒,随机将孕鼠分为溶剂对照组、BPA低剂量(10 nmol/L )组、BPA中剂量(50 nmol/L)组、BPA高剂量(300 nmol/L)组,每组6只。雄性仔鼠出生后继续饮水染毒,剂量同上,6周(PGD42)处死,取睾丸组织,计算睾丸系数;免疫组化和Western blot法检测β-cate-nin、DKK-1蛋白的表达。结果 BPA暴露组较溶剂对照组仔鼠睾丸脏器系数降低,β-catenin、DKK-1蛋白表达明显增加;免疫组化显示,β-catenin、DKK-1蛋白多位于睾丸间质细胞和精原细胞中。结论β-catenin、DKK-1可能参与了出生前后BPA暴露对子鼠雄性生殖细胞发育的影响。
