目的 观察CD151转染人脐静脉内皮细胞(HUVEC)促细胞管样结构形成,及其与整合素的相关性.方法 用脂质体转染试剂,将质粒pAAV-CD151及其突变体pAAV-CD151-AAA194-196转染HUVEC,观察转染后细胞在Matrigel胶上管样结构形成情况;免疫共沉淀法检测整合素的表达情况.结果 pAAV-CD151转染组能显著促进HUVEC管样结构形成,免疫共沉淀能检测到整合素α3、α6的表达;而CD151突变体pAAV-CD151-AAA194-196转染组促HUVEC管样结构形成能力下降,且其免疫共沉淀未能检测到整合素α3、α6的表达.结论 CD151促进HUVEC管样结构形成依赖于CD151-整合素α3/α6复合体的形成.%Objective To observe the promotive effects of pAAV-CD151 transfection into human umbilical vein endothelial cells (HUVEC)on tubular morphogenesis , and its correlation with integrins. Methods Using Liposome transfection reagent, plasmid pAAV-CD151 and pAAV-CD151-AAA194-196 mutant were transfected into HUVEC. The tubular morphogenesis on matrigel was observed. Co-immunoprecipitation assay was used to detect the expression of integrins α3 and α6. Results pAAV-CD151 transfection could significantly promote the formation of the tube-like structure of HUVEC ,and the expression of inte-grin α3 and α6 could be detected by using immunoprecipitation. pAAV-CD151-AAA194-196 transfection could not promote the formation of tube-like structure of HUVEC ,and the expression of integrins α3 and α6 could not be detected by using immunoprecipitation. Conclusion pAAV-CD151 promoted tubular morphogenesis of HUVEC depending on the formation of CD151-in-tegrin α3/α6 complex.
