目的:通过观察左旋多巴诱发的异动症( LID)大鼠脑纹状体细胞外液单胺类神经递质的代谢产物二羟基苯乙酸(DOPAC)\高香草酸(HVA)和5-羟吲哚乙酸(5-HIAA)水平,探讨首乌方(SWF)对大鼠LID的干预作用及其机制。方法:对SD大鼠进行单侧双位点注射6-羟基多巴胺造成PD模型后,灌胃投与多巴丝肼(30mg/kg/d)诱发LID,及同时灌胃中药SWF(高剂量组36g/kg/d、低剂量组27g/kg/d)。假手术组灌胃等量纯净水。给药21天中进行异常不自主运动( AIM)评分4次。之后,再次投药并在大鼠清醒自由活动状态下通过微透析技术进行纹状体细胞外液的动态采样,每30 min一管,共收集240 min。高效液相-电化学方法检测最终投药后各时间点样品中多巴胺及5-羟色胺的代谢产物DOPAC、HVA、5-HIAA的浓度。结果:SWF降低了LID大鼠的AIM评分,可明显减少DOPAC、HVA的波动性变化,降低5-HIAA浓度的升高。结论:SWF有预防及干预LID的作用,其机制可能与缓解DA受体波动性刺激及影响5-HT的浓度变化有关。%Objective:To investigate the effect of Shou Wu Fang ( SWF) and the protection mechanism by observing the concentration of DOPAC, HVA and 5-HIAA in extracellular fluids of striatum in Levodopa-induced dyskinesia ( LID) rats.Methods:Establish the PD models by unilateral perfusion of 6-OHDA in two positions in SD rats.The PD rats were administrated Madopa (30mg/kg/d) in order to induce LID models, meanwhile,they were given SWF (high dose group:27g/kg/d) or SWF ( low dose group:36 g/kg) in coadministration.Give the same amount of water to the sham group.Assess the AIM scores 4 times during 21 days.Collect the extracellular fluids of striatum by microdialysis tech-nology in free moving rats after another gavage, with 30 min/tube for 240 min.Detect the concentration of DOPAC, HVA and 5-HIAA by HPLC-ED in samples, which were metabolized by dopamine and 5-hydroxytryptamine.Re-sults:SWF reduced the AIM scores in LID rats.It could alleviate the fluctuation of DOPAC and HVA and reduce the in-crease of 5-HIAA.Conclusion:SWF has an effect on prevention and intervention of LID.It is related to the influence on the fluctuant stimulation of DA receptors and the effect on the concentration of 5-HT.
