Proteomic analysis of Abeta-associated proteins in cultured astrocytes.

It is known that β-amyloid (Aβ) is involved in accumulation of senile plaques, ROS production, impairment of endosomal/lysosomal compartment, and finally leading to neuronal cell death and Alzheimer's disease pathogenesis. Many studies investigate the relationship between Aβ and astrocytes, an important cell maintaining homeostasis in brain; however, a large amount of proteins related to Aβ and astrocytes remain elusive. The goal of this study is to identify potential Aβ-interacting proteins in astrocytes by using multi-dimensional chromatography tandem mass spectrometry (LC-MS/MS) and SEQUEST algorithm. We generated and evaluated a list of peptides corresponded to proteins associated with Aβ, and most of proteins identified can be classified into four groups: cytoskeletal organization, signaling transduction, mitochondrial energy metabolism, and gene regulation. With this powerful technology, multiple proteins can be identified in one experiment, and allow us to efficiently investigate Aβ interaction with extra- and/or intracellular proteins via an efficient way.
