Comparative proteomic analysis in fibrous root and storage root of sweetpotato

Sweetpotato [lpomoea batatas (L.) Lam.] is a representative root crop. The storage roots developed from fibrous roots of sweetpotato can be used as food and industrial materials. In this study, to understand the storage root development and differentiation of sweetpotato, we conducted comparative proteomic analysis between fibrous roots and storage roots of sweetpotato (cv. Yulmi). Total protein were extracted from each of root systems by phenol method and separated by two-dimensional elec-trophoresis (2-DE). Total 33 protein spots were differentially expressed in the fibrous root and storage root. From the result, 18 protein spots in the fibrous root and 15 protein spots in the storage root were up-regulated,respectively. We employed matrix-assisted laser desorption-time of flight (MALDI-TOF) mass spectrometry (MS) and MALDI-TOF/TOF MS analysis to identity protein spots. The up-regulated protein spots were identified as antioxidant related and protein storage related group in fibrous root and storage root, respectively. From this study, we provide the information of the regulatory proteins in sweetpotato which are involved in storage root development from fibrous root using proteomic method for the first time.
