During large-scale domestic cultivation in Yunnan and Guangxi Province, China, pseudoginseng (Panax notoginseng), a valuable traditional Chinese medicinal herb, is often attacked by Fusarium solani, causing pseudoginseng root rot disease. Recently, the protein profiles of healthy and infected roots were generated and compared. As a further step towards generating molecular markers for the study of pseudoginseng root rot pathosystem and the development of root rot disease resistant pseudoginseng cultivars, Western immnunoblot analyses were conducted with the total protein extracts from infected and healthy pseudoginseng root samples. Seven different peptide polyclonal antibodies and one polyclonal antibody (anti-Phe w I) reactive to a recombinant fungal protein (Phe w I) developed during conifer pathosystem proteomics studies were used in this Western blot study. Anti-Phe w I and four of seven anti-peptide polyclonal antibodies (16MAJ, ECP, TR2A, TR2B) reacted to proteins of healthy pseudoginseng. Fewer bands and a different banding pattern were detected in infected roots. Our results indicated that these antibodies could be used in the future as molecular tools for pseudoginseng cultivar analyses and pseudoginseng pathosystem disease response analyses.
