A method for regulating enzyme activity using the cofactor of an enzyme was developed, which was based on the apoenzyme reactivation method. This method was applied to regulate ALP (alkaline phosphatase) activity and a FIA (flow injection analysis) system was developed for evaluation of the enzyme activity. The FIA system was assembled with a syrninge pump, rotary injection valves, an immobilized enzyme column, and a UV/VIS detector. Variations in absorbance attributable to the enzyme-catalyzed reaction were monitored. Introduction of chelating agents caused a significant decrease in the enzyme activity of the column, and the activity of ALP enzyme was proportional to the concentration of zinc (II) ions.
