Application of agar bed on tobacco seed germination testing

An agar bed was developed which corresponds to a traditional paper bed,and which can be used for testing tobacco seed germination.Unprocessed seeds,primed seeds,and pelleted seeds of K326,Yun-87,and Nanjiang-3 were germinated on agar beds of different concentrations,with filter paper used as a control.When the agar concentration was 0.55％,seeds grew significantly faster than the control during early germination (p＜0.05); when it was greater than 1.65％,seedlings of naked seeds wilted on the 9th day and pelleted seeds wilted on the 10th day; when the agar concentrations were 0.8％ and 1.1％,there were no significant differences in germinative energy or germinative percentage compared with the control (p＜0.05).agar can maintain a certain amount of water used for seed germination,and needn't shower water in the whole cycle.Seeds were easy tosow and count because they cling lightly to the agar medium and did not roll as in the control.%为实现琼脂床代替纸床运用于烟草种子发芽势和发芽率的检验.以K326、云烟87和南江3号的裸种、引发种子和包衣种子为试验材料,采用梯度浓度的琼脂培养基制备发芽床,进行种子检验试验.结果表明:当琼脂浓度为0.55％时,琼脂床上的烟草种子萌发显著早于纸床(p＜0.05).当琼脂浓度大于1.65％时,裸种在9d后,包衣种在10 d后,幼苗会出现萎蔫现象；在0.8％和1.1％的琼脂床上,3个品种3种类型种子发芽势和发芽率与传统纸床检验结果均无显著差异(p＜o.05).另外与传统纸床相比,琼脂床保水性好,在整个检验周期内无需再次补水,琼脂床吸附性较好,便于烟草种子点播,在轻微震动下种子也不易滚动,所以预先摆好的苗床不易被破坏,便于计数.
