In this paper, TEP produced by two marine bacteria, SW1: pseudidiomarina homiensis and SW2:pseudoalteromonas atlantica, were isolated and characterized over time to determine theproduction rate and molecular weight of TEP. During the experiment, pure culture marinebacteria were incubated in a one-liter flask under room temperature with shaking. The bacteriawere allowed to grow in the flask for 7 days. Water samples were taken from the one-liter flaskevery day to determine the TEP content and cell number in the samples. Furthermore, thesamples were analyzed with LC-OCD to investigate the produced organic content at differentgrowing stages as well. Cell number was determined via total cell counting on peri dishes withdifferent dilution factors. The samples for TEP content determination was desalted via dialysis,and then measured with alcian blue staining method.Results of alcian blue staining showed that both marine bacteria produced TEP. Although bothmarine bacteria have a similar growth on bacteria cell number, marine bacteria SW2 producedmore TEP than SW1. The LC-OCD results showed that biopolymer, low molecular weight acidsand neutrals in the samples increased over time, indicating both high and low molecular weightsubstances were produced by marine bacteria during their growth.
