The present work discusses the development and evaluation of a novel ‘polymer-free’ microemulsion based aqua triggered in-situ (ATIS) gelling drug delivery system of sumatriptan succinate which gels rapidly on contact with minimum quantity of water. Such microemulsion compositions could therefore gel immediately on contact with wet surfaces like the nasal mucosa. Phase diagrams were plotted and ATIS gel region identified. Gelling is attributed to formation of a stiff lamellar lyotropic liquid crystalline phase as indicated by polarization microscopy. XRD spectra confirmed the formation of lyotropic liquid crystalline phases responsible for increase in viscosity on addition of water. Good correlation was obtained between viscosity and bioadhesion of ATIS gel system. The optimized ATIS gel formulation of sumatriptan succinate showed good sprayability, drug content and drug release. High retention time in rabbit nasal mucosa evaluated by gamma scintigraphic images suggest the potential of ATIS gel of Sumatriptan succinate as an effective nasal drug delivery system.
