The present work discusses the development andevaluation of a novel ‘polymer-free’ microemulsion basedaqua triggered in-situ (ATIS) gelling drug delivery systemof sumatriptan succinate which gels rapidly on contactwith minimum quantity of water. Such microemulsioncompositions could therefore gel immediately on contactwith wet surfaces like the nasal mucosa. Phase diagramswere plotted and ATIS gel region identified.Gelling isattributed to formation of a stiff lamellar lyotropic liquidcrystalline phase as indicated by polarization microscopy.XRD spectra confirmed the formation of lyotropic liquidcrystalline phases responsible for increase in viscosity onaddition of water. Good correlation was obtained betweenviscosity and bioadhesion of ATIS gel system. Theoptimized ATIS gel formulation of sumatriptan succinateshowed good sprayability, drug content and drug release.High retention time in rabbit nasal mucosa evaluated bygamma scintigraphic images suggest the potential ofATIS gel of Sumatriptan succinate as an effective nasaldrug delivery system.
