To develop a novel minocycline–loaded wound dressing, cross-linked hydrogel films were prepared using freeze-thawing method with various mixtures of polyvinyl alcohol (PVA) and chitosan. Their gel properties such as gel fraction, swelling, water vapor transmission test and morphology were investigated. The drug release from this minocycline-loaded hydrogel and in vitro protein adsorption test were then carried out. Increased chitosan concentration decreased the gel fraction, but increased the swelling ability, water vapor transmission rate and porosity of hydrogel film. However, chitosan gave no significant effect on the amounts of proteins absorbed on hydrogels and the release of drug from the hydrogels. Furtherrmore, this novel minomycinloaded wound dressing developed using freezingthawing method with PVA and chitosan gave more swellable, flexible and elastic compared to that with only PVA. Thus, this PVA/chitosan based hydrogels could be used as a potential wound dressing form with better gel property.
