Lipase immobilization on woollen cloth in the presence of transglutaminase

This paper outlines an investigation of a new method to immobilize lipase from pseudomonas fluorescens onto woollen cloth. To do this, the lipase was entrapped in cross-linked casein gel formed by transglutaminase from Streptoverticillium mobaraense (Tgase). The advantage of this technique is that lipase is immobilized via the reaction of another enzyme rather than chemical treatment. Therefore enzyme deactivation is minimized as it is due only to the physical entrapment involved. Results showed that the lipase was successfully immobilized by this method. Activity testing using a p-nitrophenyl palmitate assay showed that the lipase remains stable, despite the immobilization:The activity of the immobilized lipase was monitored continuously for more than twenty tests. Negligible activity drop was observed over these tests. Furthermore, negligible activity loss was found for this preparation after being stored in air for fifteen days. Consequently, this is a new promising stable method for immobilizing enzymes on cloths.
