Simultaneous determination of the content of Baicalin and Chlorgenic acid in Yinhuang injection by HPLC

To develop a method for simultaneously determining content of Baicalin and Chlorgenic acid in Yinhuang injection by HPLC. Method: ODS-C18 (150mm × 4.6mm, 5μm) and Gradient Elution Method. The mobile phase was methanol-0.5%KDP. Methanol-0.5%KDP (20:80) at 0.0lmin; methanol-0.5% KDP (80:20) at 0.01 min; methanol-0.5% KDP (20:80) at 11.01min. The detecting wavelength was at 316nm. Chromatographic column temperature was 40°C, and flow rate was 1/min. Results. There was a good linear relationship between the content of Chlorgenic acid and absorption area value with range of 20～60μg, recovery ratio was 99.51%, RSD=1.72%.There was a good linear relationship between the content of Baicalin and absorption area value with range of 10～30μg, recovery ratio was 99.78%, RSD=1.07%.Conclusion. The method is rapid, simple and accurate, and can be used for controlling the quality of the product.
