Antifreeze glycoprotein (AFGP) is a highly O-glycosylated protein that depresses the freezingpoint of water.This O-glycoprotein consists of an Ala-Thr-Ala glycotripeptide repeatincorporating a Gal-β-(1→3)-GalNAc (O-GalGalNAc) at the Thr residues.It has been revealedthat the O-glycosylation is an essential motif for the antifreeze activity of AFGP from manystudies performed in the past few decades.Recently we have reported that the total synthesisand functional analysis of AFGP having only O-GalNAc modification.This revealed thatAFGP can acquire both structural and functional basis of AFGP even only with the core OGalNAc.However,the functional roles of the core O-GalNAc or the O-GalGalNAc has notbeen fully understood at a molecular level.
