Fluorescent labeling is a powerful method to study biomolecules. We planned to incorporate solvatochromic fluorophore at specific lysine residue of DNA-interacting proteins, that might afford information on interactions and conformational change of proteins by change in fluorescence. Previously, we found that 6-dimethylamino-3-methyleneisoindolin-1-one 1 exhibited solvatochromic fluorescence properties;; red shift of maximum emission wavelength and decrease in fluorescence quantum yield upon increase in dielectric constant of solvent . In this study, we synthesized oligodeoxynucleotides (ODNs) 2 carrying functional group to induce 6-dimethylamino-3-methyleneisoindolin-1-one by reaction with primary amine.
