Carboxylesterase (EC 3.1.1.1) is a member of the carboxyl/cholinesterase (CCE) superfamily, which has been known to be associated with detoxification of dietary and environmental xenobiotics in insects. In this study, a novel carboxylesterase cDNA gene, HpCE19 (GenBank accession number is KC703972) was sequenced and characterized from Holotrichia parallela (Coleoptera: Motschulsky) larval. The deduced amino acid sequence showed a high identity to the reported carboxylesterase from Coleopteran insect Holotrichia oblita. Carboxylesterase activity assay in vitro showed that the recombinant HpCE19 could hydrolyze alpha-NA. Analysis of tissue-dependent expression revealed that the transcript is primarily expressed in the midgut by qRT-PCR approach. This study provides a foundation for the further research of function of H. parallela carboxylesterase HpCE19. Its biochemical and physiological functions require further investigation.
