Identification of Serum miRNA Biomarker for Early Detection of Gastric Cancer

Gastric cancer is the second leading cause of cancer death worldwide. Majority of gastric cancer are diagnosed at late stage, leading to poorer outcomes in general. Currently, there is no cost-effective method for screening of gastric cancer, making a non-invasive serum-based screening test highly desirable. To identify serum miRNA biomarker for gastric cancer, 236 Chinese with newly diagnosed gastric cancer (stage 1-4) were included in this study. Age, gender and race matched controls were selected from a high-risk cohort population with 1:1 ratio, constituting the largest sample set analyzed to date. These subjects were randomly divided into discovery set and validation set with 118 controls and 118 gastric cancers subjects each. Using a novel and enabling qPCR platform (adjacent poster), absolute expressions of 324 candidate miRNAs were quantified in all serum samples, normalized to both spike-in control RNAs and validated endogenous reference miRNAs.
