Protein molecular switches have attracted considerable attention over the past years regarding their application in affinity chromatography or as a tool for half-life extension of therapeutic proteins. Variable domains of the shark immunoglobulin antigen receptor (vNAR) are singular antibody domains which feature small size as well as extraordinary stability in comparison to conventional antibodies. Thus, pH-dependent vNAR domains with high binding affinities for their respective target protein represent suitable affinity ligands for chromatographic applications. Elution of the target protein should ideally be carried out under mild acidic conditions (6.0) in order to avoid its denaturation at lower pH.
