Asymmetrical Flow Field-Flow Fractionation (AF4) is a powerful separation and characterization technique for biological macromolecules, lipids, viruses and cell particulates. In AF4 the separation takes place in an open channel and is based on the hydrodynamic diameter or the diffusion coefficient of the respective sample components. The AF4 channel consists of two parallel walls with a thickness of 190 -500 urn. Sample components are pushed toward the semi-permeable lower channel wall by the separation field (cross flow) and transported along the channel at different flow velocities via a secondary laminar flow (channel flow).
