Serum half-life of drugs can be increased by conjugation or fusion to albumin due to both size and recycling of the molecules via the neonatal Fc receptor (FcRn). Interaction with FcRn rescues albumin, and attached therapeutic agents, from lysosomal degradation. Enhancing the interaction between albumin and FcRn was postulated to result in improved half-life of drugs, allowing tailoring to meet specific medical needs, reducing drug side effects and improving both patient compliance and quality of life. Using rational engineering strategies we have developed a new generation of safe, stable and efficacious albumins with significantly prolonged serum half-life in several animal models including cynomolgus macaques.
