Naive Library of Llama Single Domain Antibodies: An Immunization-Free Source of Specificity and Diversity

Llama single domain antibodies (llama VHHs) are the hyper-variable antigen binding domains of heavy chain-only antibodies present in llamas. Due to their specific properties, llama VHHs are excellent candidates for being used as highly specific tools and drugs. The use of single domain antibody libraries generated upon Immunization of llamas has proved to be a very valid method for the isolation of high affinity specific binders. Nevertheless, immunizations might not be the proper approach in certain cases i.e. availability of the antigen is limited, toxic antigen, immune dominant epitopes. A cost-effective alternative to bypass these issues is to use a Na?ve library of llama single domain antibodies. The generation of such library and the rapid isolation of specific llama VHHS against different target at Fairjourney Biologics will be presented.
