Lamellar Signaling Events and Disturbances of Energy Metabolism Leading to Different Forms of Laminitis

Lamellar failure occurs in the three major clinical forms of laminitis as a consequence of very different predisposing clinical situations (systemic sepsis, endocrinopathy, or preferential weight bearing). In each form of laminitis a combination of epithelial cell adhesion loss (both intercellular and cell-basement membrane) and epithelial cell stretch (likely due to cytoskeletal dysregulation) weaken the lamellar dermoepidermal attachment normally responsible for suspending the distal phalanx withinthe hoof capsule. A question of vital importance to our understanding of laminitis pathophysiology (and the development of effective therapeutic interventions) is whether this failure occurs as a result of 1) a specific cellular signaling event affecting the lamellar epithelium that is common to all forms of laminitis; or 2) different insults to epithelial cell homeostasis, which, as a result of the unique and immense physical stresses on the lamellar epithelium, have a similar end effect on this tissue's structure regardless of inciting cause.
