The aim of this research was to evaluate the effect of Glycerol on sperm motility and sperm viability of Java Barb, Barbonymus gonionotus (Bleeker, 1850) after freezing. Sperm was collected by hand stripping method and was diluted by diluent, which consisted of Fish Ringer and Glycerol. The sperm ratio to diluent was 1:3. The concentrations of Glycerol used in this research were 0%, 5%, 10%, 15%, and 20%, respectively. The sperm was frozen at -34 °C for 24 hours. According to the Dunnet test (p>0.05), 15% of Glycerol showed the highest sperm viability (68.69±5.95%), while either 10% or 15% of Glycerol showed the highest percentage of sperm motility (88.36 ± 4.77) % and (85.45 ± 10.98) %, respectively. Ten percent of Glycerol was the optimum concentration to preserve sperm motility at -34 °C for 24 hours.
