In closing, we successfully encapsulated static PLGA core NPs with a dynamic pNIPAM shell. The addition of this pNIPAM shell conferred many benefits including the ability to protect the PLGA cores from aggregating during lyophilization, the ability to modify the NPs with active targeting moieties in a concentration-dependent manner, and the ability to change size based on external environmental-based stimuli. All of these abilities will prove important in future studies focused on examining the ability of these NPs to deliver therapeutics in an arthritic-based disease model.
