Preliminary Study on Hg-Se Binding Proteins in Liver of Northern Fur Seals

In the present study,distribution of Hg and Se were determined in cytosol,and nuclear and mitochondrial fractions from liver of northern fur seals(Callorhinus ursinus).A single peak of Hg and Se in the high molecular weight substances(HMWS)of cytosol was observed at the same retention time in high performance liquid chromatography/inductively coupled plasma-mass spectrometry(HPLC/ICP-MS)analysis using GS-520HQ column.These results suggest that Se interacts with Hg,and both bind to HMW proteins in the hepatic cytosol of this species.Furthermore,Se-containing spot on gel separated with two-dimensional gel electrophoresis was detected in HMWS from northern fur seal liver.We suggest the possibility of this protein to have a pivotal role in reducing the toxicity of Hg in this species.To gain insight into the existing state of the Hg and Se in nuclear and mitochondrial fraction,they were separated by Sephadex G-75 column in extractable fractions with three extractants,sodium dodecyl sulfate;2-mercaptoethanol;2-mercaptoethanol+guanidinium thiocyanate.Mercury and Se also co-existed in HMWS of sodium dodecyl sulfate and 2-mercaptoethanol+guanidinium thiocyanate extractable fractions.
