β_2-adrenoceptor (β_2-AR) was evenly immobilized on the surface of macro-pore silica with a mild chemical coupling method through covalent bond to prepare chromatographic stationary phase. The association constant of palmatine binding to the immobilized β_2-adrenoceptor was determined by frontal analysis. The effects of mobile phase pH and ionic strength on the binding affinities of the stationary phase were also investigated. The results indicated that palmatine had one common binding site on the immobilized β_2-AR column, and the association constant was 2.93×10~4L/mol at pH 7.2 and 25°C. The interaction between palmatine and β_2-AR was mainly drived by electrostatic action.
