A laboratory study was conducted to asses the optimal conditions for polyhydroxybutyrate (PHB) production from sewage sludge and palm oil mill effluent (POME), particularly for C/N ratio using mixed culture approach. The mixed culture was prepared according to the non-limiting nutrient feeding from primarily sewage-POME inoculums. The aim was to enrich and enhance the bacterial growth, so the capability of storage polymer PHB will significant in the accumulation stage. Evaluation of PHB production was conducted under feast-famine regimes. PHA represents several PHA constituents (over 80 HAs), and PHB (or HB monomer). Sunflower oil (SO) was later used as substrate after the inoculation stage. It is observed that the highest PHB productions only achieved to 4% of dried weight (or 3.0 x 10~(-5) g/g VSS) under growth condition. This can be explained by nature of long-chain-fatty-acid (LCFA) under unsaturated fatty acid (C_(14:1) -C_(18:3)) Of SO.
