Cotyledon protoplasts of melon (cv. Cantaloup Charentais) were transformed with plasmid DNA containing neomycinphosphotransferase (nptII) and green fluorescence protein (gfp) genes, using polyethylene glycol (PEG) to induce DNA uptake. Optimal experimental conditions for plasmid concentration were analyzed. A time course study of GFP expression from protoplasts, cells, aggregates and protoplast-derived calli was performed. Results showed that 100 mu g plasmid DNA/ml of protoplasts gave the best transformation efficiency, allowing the acquisition of an average of 50 transgenic calli per million of treated protoplasts.
