Biopersistence of toner particles in rat lungs during 2 years inhalation

Wistar male rats were exposed to toner 6hrs/day, 5days/week for 2 years at 2 exposure concentration level, that is 5 and 15mg/m3. MMAD and GSD of toner particles in the chamber were 4.5μm and 1.3 respectively. Rats were sacrificed at 1 year exposure(each 10 rats) and at 2 years exposure(all remained rats). There is no difference in mortality and body(organ) weights between 2 exposure rats and controls. Lung burden of toner were determined by color imaging method. Pulmonary deposition and clearance of toner particles were mathematically estimated using the biological half time of MMMF in normal clearance condition. The clearance of high-dose exposure group was extremely prolonged at the end of exposure compared with the estimated value. This results shows that the lung burden in high-dose exposure was overloaded and induced the impairment of alveolar macrophage clearance and the excessive quantity in lung.
