The presnet study has been performed to investigate whether ketorolac, a typical anti-inflammatory drug, acts centrally by determining its effects on pain-related evoked potnetials and vigilance. Fentanly was used as a reference centrally acting analgesic. In addition, the effects of both d4ugs on subjective pain-ratings were also recorded. Fifteen human volunteers suffering from mild knee osteoarthristis participated in the experiments. The pain model used consisted of electrical tooth-pulp stimulation. The parameters. investigated were the pain-related evoked potentials.,pain-related intensity estimates. tracking performance and spontaneous EEG activity.EEG records obtained synchronously with painful stimulation were averaged by computers to improve the signal-to-noise ratio. Vigilance was measured by EEG power spectra and performance of a tracking task. Fentanyl (0.2 mg, intramuscularly) significantly reduced the vigilance, the pain ratings as well as the amplitudes of pain-related evoked potentials and increased their latencies. while ketorolac (70 mg/kg intramuscularly) did not. The results of the study suggest that ketorolac is devoid of CNS effects.
