The importance of nuclear glycoproteins, which may act as transcription factors, has been recently emphasized. It has been described that O-β-D-GlcNAc residues attached to Ser or Thr residues act as switch of the glycoprotein function. Additionally, phosphorylation (Phos) and O-β-D-GlcNAc modification of Ser/Thr residues are reciprocal, and may take place at the same or at adjacent hydroxyl moieties. Herein, we present the synthesis of different transcription factor mimics (TFM), which contain a selective-Py-γ-Py-Ind vector for interaction studies, modified by O-β-D-GlcNAc-Ser/Thr- moieties. The corresponding non-glycosylated models (Ser/Thr), and the phosphorylated analogues (Phos-Ser/Thr) have also been prepared.
