The analgesic efficiency of ketoprofen loaded in bicomponent hydrogels obtained by cross-linking a natural polysaccharide - alginate with a synthetic polymer - poly(N-isopropylacrylamide) (PNIPAAm) as sustained release dosage forms was investigated. The in vitro ketoprofen release was studied in conditions similar with the gastrointestinal environment, showing the ability of these hydrogels to achieve a sustained drug release influenced by their composition. The in vivo study by "hot plate" test performed comparatively for neat ketoprofen and ketoprofen loaded in PNIPAAm/alginate hydrogels showed the extended analgesic effect up to 24 hours for all ketoprofen-loaded hydrogels, compared with neat ketoprofen, whose antalgic effect lasts for 6 hours. The results obtained demonstrate the optimized efficiency of ketoprofen loaded in PNIPAAm/ALG hydrogels as prolonged release dosage forms.
