Introduction: The combination of polarization microscopy and staining Picrosirius red is a very efficient method to identify the stromal elements of salivary gland with and without tumors.Purpose: Identify different types of collagen in the normal and tumoral stroma of the salivary glands by combining the technique of Picrosirius red with polarization microscopy.Material and Methods: We used paraffin-embedded biopsias of patients with pleo-morphic adenoma (n= 5), adenoid cystic carcinoma (n =4) and epithelial-myoepithelial carcinoma (n =1) The histological sections were stained with H/E, Picrosirius red and observed under a polarization microscope NIKON MF 100POL fishplates.Results: Examination of the birefringence of the stroma of pleomorphic adenoma showed the presence of type in collagen fibers with a range from green-yellow color to green. Epithelial-myoepithelial carcinoma and adenoid cystic carcinoma showed a predominance of type I collagen, with birefringence from orange yellow to orange and red.Conclusions: The method of Picrosirius red staining and visualization with polarized light microscopy is a simple and very efficient technique to identify, locate and characterize different types of collagen (I and IE) in easy and accurate way. The results obtained with the above techniques allowed the identification of different stromal elements according to the salivary gland tumors studied, it would allow in future to provide a better diagnosis and patient management.
