The Klason lignin and benzene-ethanol extracts content of 78 Triploid of Populus Tomentosa samples were analyzed according to traditional method firstly and then measured by Near Infrared Spectroscopy. After the spectra were treated with second derivative and linear offset subtraction, then the NIR model was built with partial least-squares statistical analysis and full cross validation. The coefficients of correlation(R) of calibration and validation for lignin were 0.8883 and 0.8634; the standard error of calibration (SEC) and the standard error of prediction (SEP) were 0.612 and 0.944, respectively. For benzene-ethanol extracts the coefficients of correlation of calibration and validation were 0.9007 and 0.8857, while the SEC and SEP were 0.39 and 0.603, respectively. It was confirmed that NIR analysis is a reliable testing predictor of lignin and benzene-ethanol extracts content of the poplar wood.
