>200,000 theoretical plates were achieved for <5 minute long CZE separations for kinase insulin receptor peptide standard. <10 attomole detection limits were achieved for some peptides from four standard protein tryptic digest in CZE-MS with LTQ Orbitrap Velos. 2 minute separation time window was sufficient for exhaustive analysis of four standard protein tryptic digest. Analysis of peptide mixtures of higher complexity can be accomplished by increasing separation time window or the number of technical replicates. We demonstrated successful CIEF-MS peptide separations, based solely on chemical mobilization and without the need of gravity assistance.
