Development of a top-down approach for Staphylococcus aureus enterotoxins A and B absolute quantification

In summary, we have evaluated top-down quantification of toxins, among them the SEA and SEB enterotoxins. Combination of intact high resolution mass spectrometry analysis with on beads immunocapture provide a rapid (30 minutes sample preparation, less than total 4 hour run time for a batch of ～10 samples) and sensitive (down to ng injected on column) detection tools. Addition of PSAQ standards directly into the samples to be analyzed will allow a highly accurate quantification of SEA/SEB. This work could be extended to the other Staphylococcal enterotoxins.
