Metformin, a commonly prescribed drug for type 2 diabetes, is also thought to play a role in cancer prevention acting as a calorierestriction mimetic. Previously, we reported that calorie restriction significantly reduced mammary tumor incidence in a mouse model of breast cancer, while diet-induced obesity enhanced tumor development. An ongoing study is focused on the effect metformin has on mammary tumor development in this mouse model. However, metformin concentration in serum samples from treated mice or human subjects is not well characterized, which is likely due to the lack of internal standard and a reliable detection method for metformin measurement. Here, we seek to quantify metformin concentrations in the serum samples of metformin-treated mice via HILIC-SID-SRM-MS. 61st ASMS
