Evaluated four different methodologies using combination of HRAM MS, HILIC, UHPLC, Nanospray/nano flow. Method 1 (HILIC, isocratic): (1)Direct transfer to 1D HILIC chromatography was not successful; Method 2 (HILIC, forward flush): (2) Acceptable result but too sensitive to aqueous content. Method 3 (Phenyl Hexyl, UHPLC): (1) Most promising. No carryover. PLs under control with backflush; Method 4 (C18, Nanocolumn and Easy spray source): (2) Good precision and accuracy. But can't do column backflush. Handling of phospholipids are still a challenge.
