AMCA-X was used to modify model peptides and protein digests at the N-terminus to attached a chromophore to enable efficient UVPD at 351 nm. UVPD of AMCA-X modified peptides resulted in conventional b/y type ions, but the AMCA-containing b ion series could be selectively annihilated while the y ions survived stable products upon multiple UV pulses. This results in UVPD spectra that exhibit only the y ion series and thus are well suited for de novo sequencing.
